Throughout the world, infections with the Babesia and Theileria parasites often result in economically significant clinical disease in cattle. We conducted a longitudinal survey of Babesia and Theileria infections in cattle from the Polonnaruwa (n?=?75; dry zone) and Nuwara Eliya (n?=?161; wet zone) districts of Sri Lanka. DNA from blood samples collected in June, September, and December 2014 and March 2015 was screened for Babesia bovis, Babesia bigemina, Theileria annulata and Theileria orientalis using specific polymerase chain reactions (PCRs). Additionally, serum samples collected from the animals were screened using enzyme-linked immunosorbent assays (ELISAs) to detect B. bovis- and B. bigemina-specific antibodies. All of the animals surveyed in...

An epidemiological survey on a Theileria parasite infection of cattle in Northeast China was carried out using allele-specific PCR and DNA sequence analysis of the major piroplasm surface protein (MPSP) gene. The results showed that 14 of 104 blood samples were positive for Theileria by PCR. Among the positive cases, co-infection with various combinations of C- and I-type parasites was detected in 12 samples; no B- and Thai-type parasites were detected by allele-specific PCR. Phylogenetic analysis based on the MPSP gene sequences revealed that Theileria parasites with the MPSP types 1, 2, and 4 were distributed in Northeast China.

Evaluation of inhibitory effect of methylene blue against Babesia and Theileria parasites OAK
Tuvshintulga, B.; Sivakumar, T.; Salama, A.A.; Yokoyama, N.; Igarashi, I; 五十嵐, 郁男; 横山, 直明.
Methylene blue, the first fully synthetic drug, has been widely used in medical treatments. A few decades ago, this drug was used as an antimalarial agent. In this study, the in vitro inhibitory effect of methylene blue on Babesia bovis, B. bigemina, B. caballi, and Theileria equi (B. equi) and the in vivo inhibitory effect on B. microti were evaluated. Methylene blue significantly inhibited the growth of B. bovis, B. caballi, and T. equi at a 0.1 μM concentration, while B. bigemina was significantly inhibited at 0.01 μM, on day 3 of cultivation. The half maximal inhibitory concentrations (IC50) of methylene blue against B. bovis, B. bigemina, B. caballi, and T. equi were calculated as 0.83±0.02, 0.68±0.09, 0.54±0.14, and 0.49±0.06 μM, respectively. The...

Evaluation of inhibitory effects of dipyridamole against bovine and equine piroplasmosis OAK
Tuvshintulga, B.; Sivakumar, T.; Yokoyama, N.; Igarashi, I..
Dipyridamole, an antiplatelet drug used for the secondary prevention and treatment of stroke, also has antiplasmodial activity and enhances the activity of chloroquine. We evaluated the inhibitory effects of dipyridamole on the growths of Babesia and Theileria parasites. The growth of B. bovis and T. equi was significantly inhibited at a 25 μM concentration of dipyridamole, while B. bigemina and B. caballi were significantly inhibited at 50 μM and 1 μM concentrations of dipyridamole, respectively, on day 3 of cultivation. The half maximal inhibitory concentration (IC50) of dipyridamole was calculated at 39.5±4.5, 26.3±10.9, 13.2±3.6, and 23.5±0.5 μM on the growth of B. bovis, B. bigemina, B. caballi, and T. equi, respectively. In a viability assay, B....
